Principle of the Assay::The Human IL-32 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IL-32 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human IL-32 coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-32 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IL-32 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-32 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: IL-32alpha is the shortest and most abundant of four potential splice variants of the proinflammatory cytokine IL-32 (previously called NK4) with a predicted unmodified size of 15 kDa. Potential modifications include myristoylation and N-glycosylation. The IL-32alpha shows increased potency at inducing CXCL2/MIP-2 and CXCL8 expression in PBMC relative to uncleaved IL-32alpha. Transfected IL-32alpha was more likely to be cellassociated as compared to IL-32beta, suggesting an intracellular function. IL-32 is induced by mitogens in peripheral lymphocytes, by IFNgamma in epithelial cells, or by IL-12 with IL-18 in NK cells and in turn induces cytokine expression
